RESUMO

Asthma is an inflammatory lung disease that affects more women than men in adulthood. Clinical evidence shows that hormonal fluctuation during the menstrual cycle and menopause are related to increased asthma severity in women. Considering that life expectancy has increased and that most women now undergo menopause, strategies to prevent the worsening of asthma symptoms are particularly important. A recent study from our group showed that re-exposure of ovariectomised allergic mice to antigen (ovalbumin) leads to an exacerbation of lung inflammation that is similar to clinical conditions. However, little is known about the role of probiotics in the prevention of asthma exacerbations during the menstrual cycle or menopause. Thus, our objective was to evaluate the effects of supplementation with kefir, a popular fermented dairy beverage, as a preventive strategy for modulating allergic disease. The results show that the preventive kefir administration decreases the influx of inflammatory cells in the airways and exacerbates the production of mucus and the interleukin 13 cytokine. Additionally, kefir changes macrophage polarisation by decreasing the number of M2 macrophages, as shown by RT-PCR assay. Thus, kefir is a functional food that potentially prevents allergic airway inflammation exacerbations in ovariectomised mice.

Allergic contact dermatitis (ACD) is a common allergic skin disease that affects individuals subjected to different antigen exposure conditions and significantly impacts the quality of life of those affected. Numerous studies have demonstrated that probiotics suppress inflammation through immunomodulatory effects. In this study, we aimed to evaluate the effect of the probiotic Bifidobacterium longum 51A as a preventive treatment for ACD using an oxazolone-induced murine model. We demonstrated that B. longum 51A exerted a prophylactic effect on oxazolone-induced ACD-like skin inflammation via reductions in ear and dermal thickness and leucocyte infiltration. The administration of inactivated B. longum 51A did not affect oxazolone-induced ACD-like skin inflammation, suggesting that the bacteria must be alive to be effective. Given that B. longum 51A is an acetate producer, we treated mice with acetate intraperitoneally, which also prevented ear and dermal thickening. Moreover, the tissue levels of the inflammatory cytokines and chemokines interleukin (IL)-10, IL-33, tumour necrosis factor-α, chemokine (C-C motif) ligand 2/monocyte chemoattractant protein-1 and chemokine (C-C motif) ligand 5/RANTES were significantly reduced after probiotic treatment, but only IL-33 and IL-10 were reduced when the mice were treated with acetate. These results show that B. longum 51A exerted a potential prophylactic effect on skin inflammation and that acetate represents one potential mechanism. However, other factors are likely involved since these two treatments do not yield the same results.

This study aims to determine the antitumor potential of cashew gum in vitro and in vivo. The cashew gum (CG) structure is similar to already showed in literature. The cytotoxicity effect of CG was performed by MTT assay, and B16-F10 melanoma model was used to evaluate antitumor effect. The tumor inhibition was calculated based on tumor weight. Hematological, histopathological, FTIR, oxidative stress and Western Blot analysis were performed to elucidate the mechanism of inhibition and toxic effects. As results, CG did not demonstrate cytotoxicity in vitro, however showed a significant tumor inhibition in vivo, with about 36.9 to 43% of reduction in tumor mass, with no toxicity to organs. Animals treated with CG did not show toxicity in normal tissues, FTIR spectrum and oxidative stress analysis of the tumor tissue indicated that CG cause tumor inhibition with the presence of apoptosis morphotype cells, without alterations in the levels of antioxidants components. In addition, it was observed that CG reduced the expression of γH2AX without changing the expression of caspase-3. With this, we can suggest that this polymer can assist in the anticancer activity and/or decrease the side effects of standard drugs used in treatment of cancer.

BACKGROUND: Photodynamic therapy (PDT) is an anticancer therapy that associates the photosensitizer (PS), oxygen and light to destroy cancer cells. Methylene blue (MB) is considered a second generation phenothiazine dye with excellent photochemical properties. AIM: To evaluate whether MB-mediated PDT can induce oxidative stress and inflammation, therefore, interfering tumor growth. MATERIALS AND METHODS: The study was conducted on Wistar rats transplanted with Walker 256 carcinosarcoma (W256). The proinflammatory interleukins levels (IL-1ß, IL-6, IL-10, TNF-α) were determined by ELISA, mRNA expression of COX-1, COX-2, iNOS and eNOS by RT-PCR, lipid peroxidation was measured by the TBARS method. Moreover, myeloperoxidase (MPO) activity in neutrophils was determined by MPO activity assay. All indices mentioned above were determined in tumor tissue. Kaplan - Meier and Gehan - Breslow - Wilcoxon tests were used for survival analysis. RESULTS: We found that the treatment of W256 with 0.1% MB + 1 J/cm2 provoked a significant increase in the interleukins levels (IL-1ß, IL-6, IL-10, TNF-α), prostaglandin E2, the mRNA expression of COX-2, iNOS, lipid peroxidation and MPO activity in tumor tissue, which were statistically different (p < 0.05) compared to other experimental and control groups. The results of the estimation of survival curves show a greater probability of survival in 0.1% MB + 1 J/cm2 (total energy dose =142.8 J/cm2) treated group. CONCLUSION: Our results suggest that treatment of W256 with 0.1% MB + 1 J/cm2 was able to promote cytotoxic effects in tumor tissue by the generation of reactive oxygen species causing inflammation and thus interfering in the tumor growth.

Blends of rice waste streams were submitted to co-gasification assays. The resulting chars (G1C and G2C) were characterized and used in Cr(III) removal assays from a synthetic solution. A Commercial Activated Carbon (CAC) was used for comparison purposes. The chars were non-porous materials mainly composed by ashes (68.3-92.6% w/w). The influences of adsorbent loading (solid/liquid ratio - S/L) and initial pH in Cr(III) removal were tested. G2C at a S/L of 5 mg L-1 and an initial pH of 4.50 presented an uptake capacity significantly higher than CAC (7.29 and 2.59 mg g-1, respectively). G2C was used in Cr(III) removal assays from an industrial wastewater with Cr(III) concentrations of 50, 100 and 200 mg L-1. Cr(III) removal by precipitation (uptake capacity ranging from 11.1 to 14.9 mg g-1) was more effective in G2C, while adsorption (uptake capacity of 16.1 mg g-1) was the main removal mechanism in CAC.

Allergic asthma is a chronic disease mainly characterised by eosinophil inflammation and airway remodelling. Many studies have shown that the gut microbiota of allergic individuals differs from that of non-allergic individuals. Although high levels of bifidobacteria have been associated with healthy persons, Bifidobacterium adolescentis ATCC 15703, a gut bacteria, has been associated with allergic individuals in some clinical studies. The relationship between B. adolescentis ATCC 15703 and asthma or allergies has not been well elucidated, and its effect may be dependent on the host's genetic profile or disease state. To elucidate this question, we evaluated the role of preventive B. adolescentis ATCC 15703 treatment on experimental allergic airway inflammation in two genetically different mouse strains, Balb/c and C57BL/6 (B6). Balb/c mice display a greater predisposition to develop allergic responses than B6 mice. Oral preventive treatment with B. adolescentis ATCC 15703 modulated experimental allergic airway inflammation, specifically in Balb/c mice, which showed decreased levels of eosinophils in the airway. B6 mice did not exhibit any significant alterations in eosinophils but showed an increased influx of total leukocytes and neutrophils into the airway. The mechanism underlying the beneficial effects of these bacteria in experimental allergic mice may involve products of bacteria metabolism, as dead bacteria did not mimic the ability of live B. adolescentis ATCC 15703 to attenuate the influx of eosinophils into the airway. To conclude, preventive oral B. adolescentis ATCC 15703 treatment can attenuate the major characteristic of allergic asthma, eosinophil airway influx, in Balb/c but not B6 mice. These results suggest that oral treatment with this specific live bacterial strain may have therapeutic potential for the treatment of allergic airway disease, although its effect is mouse-strain-dependent.

OBJECTIVES: To demonstrate a study design that could be useful in low-resource and violent urban settings and to estimate the prevalence of child violence exposure (at home, community, and school) and child mental health problems in a low-income medium-size city. METHODS: The Itaboraí Youth Study is a Norway-Brazil collaborative longitudinal study conducted in Itaboraí city (n = 1409, 6-15 year olds). A 3-stage probabilistic sampling plan (random selection of census units, eligible households, and target child) generated sampling weights that were used to obtain estimates of population prevalence rates. RESULTS: Study strengths include previous pilot study and focus groups (testing procedures and comprehension of questionnaire items), longitudinal design (2 assessment periods with a mean interval of 12.9 months), high response rate (>80%), use of standardized instruments, different informants (mother and adolescent), face-to-face interviews to avoid errors due to the high frequency of low-educated respondents, and information gathered on a variety of potential predictors and protective factors. Children and adolescents presented relevant levels of violence exposure and clinical mental health problems. CONCLUSIONS: Prevalence estimates are probably valid to other Brazilian low-income medium-size cities due to similarities in terms of precarious living conditions. Described study methods could be useful in other poor and violent world regions.

RESUMO

Conductance was measured in two different double stranded DNA (both with 20 bases), the more conducting poly(dG)-poly(dC) (ds-DNAc) and the less conducting poly(dA)-poly(dT) (ds-DNAi), by means of Electrochemical Capacitance Spectroscopy (ECS). The use of the ECS approach, exemplified herein with DNA nanowires, is equally a suitable and time-dependent advantageous alternative for conductance measurement of molecular systems, additionally allowing better understanding of the alignment existing between molecular scale conductance and electron transfer rate.

OBJECTIVE: To verify the association between violence and alcohol dependence syndrome in sample populations. METHOD: Population-wide survey with multistage probabilistic sample. 3,744 individuals of both genders, aged from 15 to 75 years, were interviewed from the cities of São Paulo and Rio de Janeiro using the Composite International Diagnostic Interview (CIDI 2.1). RESULTS: In both cities, alcohol dependence was associated with the male gender, having suffered violence related to criminality, and having suffered familial violence. In both cities, urban violence, in more than 50% of cases, and familial violence, in more than 90% of cases, preceded alcohol dependence. The reoccurrence of traumatic events occurred in more than half of individuals dependent on alcohol. In São Paulo, having been diagnosed with PTSD is associated with violence revictimization (P = 0.014; Odds = 3.33). CONCLUSION: Alcohol dependence syndrome is complexly related to urban and familial violence in the general population. Violence frequently precedes alcoholism, but this relationship is dependent on residence and traumatic events. This vicious cycle contributes to perpetuating the high rates of alcoholism and violence in the cities. Politicians ordering the reduction of violence in the large metropolises can, potentially, reduce alcoholism and contribute to the break of this cycle.

Objetivo. Discutir os resultados acerca dos efeitos do uso da eritropoietina humana recombinante (rHuEPO) sobre o desempenho físico, bem como relatar os efeitos adversos decorrentes desta prática, correlacionando os achados de estudos experimentais e clínicos. Métodos. Foram selecionados artigos científicos publicados nas bases de dados PubMed e SciELO, no período de 1985‐2013, utilizando as seguintes palavras‐chaves: eritropoietina/erythropoietin, desempenho atlético/athletic performance, resistência física/physical endurance, efeitos adversos/adverse effects e doping nos esportes/doping in sports. Resultados. Todos os estudos (n = 10) encontraram melhora nas variáveis de consumo máximo de oxigênio (VO2máx) e tempo de exaustão em humanos, utilizando diferentes protocolos de dosagens, que podem variar de 50‐60 UI/kg nas primeiras semanas, com redução ao longo do tratamento. Dentre as reações adversas mais frequentes estão os acidentes trombovasculares, deficiência de ferro e hipertensão arterial sistêmica. Conclusão. O tratamento com rHuEPO em diferentes doses e períodos pode potencializar o desempenho físico de humanos, em virtude dos diferentes efeitos gerados, incluindo aumento no transporte de O2, redução das concentrações de lactato sanguíneo, aumento das concentrações de ácidos graxos livres no sangue e do glicogênio muscular (AU)

Discutir los resultados relativos a los efectos del uso de la eritropoyetina humana recombinante (rHuEPO), sobre el rendimiento físico, así como los efectos adversos de su uso, así como la relación de los resultados de estudios experimentales y clínicos. Métodos. Se seleccionaron los artículos científicos publicados en revistas incluidas en PubMed y SciELO, en el período de 1985‐2013, usando las siguientes palabras clave: eritropoietina/erythropoietin, desempenho atlético/athletic performance, resistência física/physical endurance, efeitos adversos/adverse effects e doping nos esportes/doping in sports. Resultados. Todos los artículos (n = 10), en relación con la influencia del tratamiento con rHuEPO en el rendimiento deportivo, mostraron una mejoría en las variables de consumo máximo de oxígeno (VO2máx) y el tiempo hasta el agotamiento en seres humanos, utilizando diferentes protocolos de dosis, que oscilan desde 50‐60 UI/kg en las primeras semanas con reducción progresiva durante el tratamiento. Entre los efectos adversos más frecuentes se encuentran los accidentes vasculares trombóticos, la ferropenia y la hipertensión arterial. Conclusión. El tratamiento con dosis de rHuEPO y en diferentes períodos puede mejorar el rendimiento físico en los seres humanos, debido a los diferentes efectos generados, incluyendo el aumento del transporte de O2, la reducción de las concentraciones de lactato en sangre, aumento de las concentraciones de ácidos grasos libres en la sangre y glucógeno muscular (AU)


Objective. Discuss the influence of rHuEPO on physical performance, as well as the adverse effects of this practice, exposing results from experimental and clinical studies. Methods. Were selected articles published in the basis, PubMed and SciELO databases, in the period 1985‐2013, using the following keywords eritropoetina/erythropoietin, desempenho atlético/athletic performance, resistência física/physical endurance, efeitos adversos/adverse effects e doping nos esportes/doping in sports. Results. All articles (n = 10) related to the influence of treatment with rHuEPO on sports performance, found improvement in the variables of maximal oxygen uptake (VO2max) and time to exhaustion in humans, using different protocols dosages between 50 to 60 IU/kg during the first weeks with reduced throughout treatment. Among the most common adverse effects are the thrombovascular accidents, iron deficiency and hypertension. Conclusion. Treatment with rHuEPO doses and at different periods, can enhance physical performance in humans, because of the different generated effects, including increased O2 transport, reduction of blood lactate concentrations, increased concentrations of free fatty acids in the blood and muscle glycogen (AU)

Herbal medicines with anthelmintic effects are alternatives for the sustainable control and prevention of disease caused by gastrointestinal parasites. The nanoencapsulation of essential oils has been proposed to enhance the absorption of their constituents and improve their efficacy. The present study aimed to evaluate the efficacy of free and nanoencapsulated Eucalyptus citriodora essential oil (EcEO) on the control of gastrointestinal nematodes of small ruminants in vitro and in vivo. Chitosan was used as a matrix for the formulation of a nanoemulsion. Chromatographic and physico-chemical analyses of EcEO were performed. Egg hatch (EHT) and larval development (LDT) tests were conducted to evaluate the effectiveness of nanoencapsulated and free EcEO on the eggs and larvae of Haemonchus contortus. Acute toxicity of free and nanoencapsulated EcEO was evaluated using mice. Finally, nanoencapsulated EcEO efficacy on the control of gastrointestinal nematodes was calculated by fecal egg count reduction test (FECRT) treating 30 sheep naturally infected with 250 mg/kg of free and nanoencapsulated EcEO. In vitro tests were analyzed by an analysis of variance (ANOVA) followed by comparison with the Tukey test. The efficacy of FECRT was calculated by the BootStreet program through arithmetic average, using the formula 100 (1-XT/XC). To compare the differences between epg, the data were transformed to log(x+1) and subjected to an ANOVA to compare the significant differences between groups by Tukey's. The level of significance was P<0.05. The free (4 mg/ml concentration) and nanoencapsulated (2mg/ml concentration) EcEO inhibited larvae hatching by 97.2% and 92.8%, respectively. Free and nanoencapsulated EcEO at 8 mg/ml inhibited larval development by 99.8% and 98.1%, respectively. In the acute toxicity test, the LD10 and LD50 of free EcEO was 1999 and 2653 mg/kg, respectively, while the LD10 and LD50 of nanoencapsulated EcEO was 1121 and 1681 mg/kg, respectively. Nanoencapsulated and free EcEO reduced FEC similarly by 40.5% and 55.9%, respectively at 10 days post-treatment. Nanoencapsulated EcEO did not obtain the expected efficacy in vivo.

Objectives. To verify the relationship between postural changes, osteoarthritis, and bone mineral density in women with osteoporosis in a research group of participants in this area. Methods. Seventy four Brazilian women aged 50 years or older with osteoporosis sent to Clinical Hospital were assessed. After anamnesis the subjects were evaluated in posture with a postural framework. The experimental protocol consisted of two groups: control group (without postural changes) and study group (with postural changes). Results. The age average in the study group was greater than in the control group. In the joint changes, 65 subjects showed osteoarthritis in some part of the body. In the whole group assessed, 19 showed change in posture and all of them have osteoarthritis too. Conclusion. This research showed that posture changes are direct relationship with advanced age. It concluded that posture assessment is essential to prescribe the most objective and efficient exercise program, with understanding of the biomechanical incidence forces, but not forgetting to strengthen the muscles to maintain the corporal posture(AU)


Objetivos. Verificar la relación entre los cambios posturales, la osteoartritis y la densidad mineral ósea en mujeres con osteoporosis en un grupo de investigación de los participantes en esta área. Métodos. Se evaluó a setenta y cuatro mujeres brasileñas de 50 años o mayores con osteoporosis en el Hospital Clínico. Después de anamnesis las mujeres fueron evaluadas respecto a la postura con una cuadrícula postural. El protocolo experimental consistió en dos grupos: grupo control (sin cambios posturales) y el grupo de estudio (con los cambios posturales). Resultados. El promedio de edad en el grupo de estudio fue mayor que en el grupo control. En los cambios en las articulaciones, 65 mujeres mostraron osteoartritis en alguna parte del cuerpo. En todo el grupo evaluado, 19 mostraron un cambio en la postura y todas ellas tenían osteoartritis también. Conclusión. Esta investigación mostró que los cambios posturales tienen relación directa con la edad avanzada. Concluye que la evaluación de la postura es esencial para prescribir el programa de ejercicios más objetivo y eficiente, con la comprensión de las fuerzas biomecánicas de incidencia, pero sin olvidar fortalecer los músculos para mantener la postura corporal(AU)

Este trabalho teve como objetivo avaliar a vida útil pós-colheita de folhas de Capuchinha (Tropaeolum majus L.) embaladas com filme PVC de baixa densidade e proteínas nas concentrações de 1, 3 e 5 por cento. O experimento foi conduzido no Laboratório de Química e Bioquímica do Centro de Ciências Agrárias (CCA) da Universidade Federal da Paraíba (UFPB) Areia - PB. As folhas de Capuchinha colhidas no CCA foram levadas imediatamente ao Laboratório de Química e Bioquímica, para pré-seleção, desinfestação em água clorada contendo 100mg.L-1 de cloro ativo, e seca à temperatura ambiente. Após o controle fitossanitário, as folhas foram submetidas aos seguintes tratamentos: solução de proteína nas concentrações de 1, 3 e 5 por cento, filme PVC de baixa densidade e a testemunha sem recobrimento. As folhas foram armazenadas durante cinco dias em temperatura média de 26 - 29ºC e umidade relativa média de 59,5 - 71,5 por cento e outra parte foi armazenada em câmara fria com temperatura média de 12 ± 0,5 ºC e umidade relativa média de 95 ± 3 por cento. Durante o período de armazenamento as folhas foram avaliadas quanto à perda de massa fresca, determinação de ácido ascórbico, sólidos solúveis e acidez titulável. O armazenamento durante quatro dias sob refrigeração a temperatura média de 12 ± 0,5ºC associada ao recobrimento com filme PVC mostrou melhores resultados na conservação da vida útil pós-colheita das folhas de Capuchinha.


This study aimed to evaluate the shelf-life of Capuchin sheets (Tropaeolum majus L.) packed with plastic wrap and low density proteins at concentrations of 1, 3 and 5 percent. The experiment was conducted at the Laboratório de Química e Bioquímica and the Centro de Ciências Agrárias (CCA) of the Universidade Federal da Paraíba (UFPB) Areia - PB. Capuchin leaves harvested in the CCA were taken immediately to the Laboratório de Química e Bioquímica, for pre-screening, disinfection in chlorinated water containing 100mg.L-1 of active chlorine, and dried at room temperature. After spraying, the leaves were treated as follows: protein solution at concentrations of 1, 3 and 5 percent, low density plastic wrap and uncoated witness. The leaves were stored for five days in average temperature from 26 to 29ºC and relative humidity from 59.5 to 71.5 percent and another part was stored in cold with average temperature of 12 ± 0.5ºC and relative humidity of 95 ± 3 percent. During the storage period the leaves were evaluated for weight loss, determination of ascorbic acid, soluble solids and titratable acidity. Storage for four days under refrigeration at an average temperature of 12 ± 0.5ºC associated with PVC film coating showed better results in the conservation of shelf-life of Capuchin sheets.

This article reports the anti-inflammatory effect of Blutaparon portulacoides (B. portulacoides), specifically the ethanolic extract of its aerial parts, on the edema formation and leukocyte influx caused by Bothrops jararacussu (B. jararacussu) snake venom and Bothropstoxin-I and II (BthTX-I and II) isolated from this venom as an alternative treatment for Bothrops snakebites. The anti-inflammatory effect of B. portulacoides ethanolic extract was compared with an animal group pretreated with dexamethasone. B. portulacoides ethanolic extract significantly inhibited paw edema induced by B. jararacussu venom and by BthTX-I and II. Also, results demonstrated that the extract caused a reduction of the leukocyte influx induced by BthTX-I. However, the extract was not capable of inhibiting the leukocyte influx induced by the venom and by BthTX-II. In conclusion, these results suggest that the ethanolic extract of this plant possess components able to inhibit or inactivate toxins present in B. jararacussu venom, including its myotoxins, responsible for the edema formation. However, the leukocyte migration caused by the venom and BthTX-II was not inhibited by the plant, probably due to the different mechanisms involved in the edema formation and leukocyte influx. This is the first report of B. portulacoides extract as anti-inflammatory against snake venoms and isolated toxins.(AU)

Venom of the South American rattlesnake, Crotalus durissus terrificus (Cdt), presents myotoxic and neurotoxic outcomes, but reports on its effects on the liver are scarce. This study examined the hepatotoxicity resulting from Cdt venom administration (100, 200 and 300 miug/kg) in male Wistar rats. Animais were studies at 3, 9 and 12 hours after venom injection. The hepatotoxicity was assessed through serum levels of aspartate aminotransferase (AST), alanine aminotransferase (ALT), alkaline phosphatase (AP), gamma glutamyl transferase (GGT), bilirrubin and also by histopathological evaluation. All the different concentrations of Cdt venom resulted in increased levels of hepatic enzymes, when compared with the control group, except for the 100 miug/kg dose, which presented normal levels at 9 and 12 hours after venom administration. Bilirrubin levels remained unchanged by Cdt venom. Histological analysis revealed endothelial damage, inflammatory cell infiltration, as well as sinusoidal and portal congestion. Based on these observations, we may conclude that Cdt venom causes dose- and time-dependent hepatic damage in rats, characterized by elevated hepatic enzyme levels and histological alterations
